In small and fragmented populations, genetic diversity may be reduced owing to increased levels of drift and inbreeding. This reduced diversity is often associated with decreased fitness and a higher threat of extinction. However, it is difficult to determine when a population has low diversity except in a comparative context. We assessed genetic variability in the critically endangered Cross River gorilla (Gorilla gorilla diehli), a small and fragmented population, using 11 autosomal microsatellite loci. We show that levels of diversity in the Cross River population are not evenly distributed across the three genetically identified subpopulations, and that one centrally located subpopulation has higher levels of variability than the others. All measures of genetic variability in the Cross River population were comparable to those of the similarly small mountain gorilla (G. beringei beringei) populations (Bwindi and Virunga). However, for some measures both the Cross River and mountain gorilla populations show lower levels of diversity than a sample from a large, continuous western gorilla population (Mondika, G. gorilla gorilla). Finally, we tested for the genetic signature of a bottleneck in each of the four populations. Only Cross River showed strong evidence of a reduction in population size, suggesting that the reduction in size of this population was more recent or abrupt than in the two mountain gorilla populations. These results emphasize the need for maintaining connectivity in fragmented populations and highlight the importance of allowing small populations to expand.  相似文献

 研究残存居群的数量性状分化及其地方适应性有助于了解不同居群对环境的适应性并制定相应的保育策略。本研究采用同园实验,利用随机区 组设计,对中华水韭(Isoetes s inensis)3个居群的9种优势基因型的数量性状进行巢式方差分析,并采用Bayesian方法计算等位酶(FST)和 数量性状(QST)的居群分化值。结果表明,在测量的14个数量性状中,有10个性状在居群间有显著性差异,仅有3个性状在居群内有显著性差 异;多重比较表明其中5个性状的平均值均以松阳居群最高,休宁居群最低;建德居群的大孢子囊的性状和植株高度的平均值最高,休宁居群最 低;推测这可能是与奠基者效应、物种间竞争及中华水韭在自然生境下形成的营养生长和生殖生长之间的权衡有关。对居群数量性状分化的QST 值和等位酶分化的FST值分析比较发现,在假设为自交系时有8个性状的QST值均大于FST值,表明存在明显的地方适应性,居群间个体的混合存 在潜在的远交衰退的风险,所以不宜采用居群间相互移植个体的方法来进行中华水韭居群的遗传复壮。对休宁和松阳两个居群的保育应采用提 高居群内基因流和改善生境环境的方法,促进居群内的遗传复壮;对建德居群应尽量减少人为干扰,宜建立特定生境的保护小区。  相似文献

The giant panda is a global symbol of wildlife conservation that is threatened by historic and current habitat loss. Despite a great deal of research on the physiology, reproductive biology, and diet of pandas in the wild and in captivity, there is little information on wild panda mortality. Here we integrate previously unavailable data on the mortality of wild pandas. We report on three recent phases of panda mortality: deaths due to bamboo flowering in the 1970s and 1980s, surprisingly extensive poaching in the 1980s and 1990s, and a parasitic infection over the past few years. Our analyses suggest that the current most significant threat to wild panda survival is disease due to extraintestinal migration (visceral larval migrans) by an ascarid nematode. We demonstrate that the probability of death of wild pandas being caused by this disease increased significantly between 1971 and 2005 and discuss the possible factors leading to the emergence of this disease. Vitexrotundifolia L.is an important plant species used in traditional Chinese medicine.For its efficient use and conservation,genetic diversity and clonal variation of V.rotundifolia populations in China were investigated using inter-simple sequence repeat markers.Fourteen natural populations were included to estimate genetic diversity,and a large population with 135 individuals was used to analyze clonal variation and fine-scale spatial genetic structure.The overall genetic diversity (GD) of V.rotundifolia populations in China was moderate (GD=0.190),with about 40% within-population variation.Across all populations surveyed,the average within-population diversity was moderate (P = 22.6%; GD = 0.086).A relatively high genetic differentiation (Gst=0.587)among populations was detected based on the analysis of molecular variance data.Such characteristics of V.rotundifolia are likely attributed to its sexual/asexual reproduction and limited gene flow.The genotypic diversity (D=0.992) was greater than the average values of a clonal plant,indicating its significant reproduction through seedlings.Spatial autocorrelation analysis showed a clear within-population structure with gene clusters of approximately 20 m.Genetic diversity patterns of V.rotundifolia in China provide a useful guide for its efficient use and conservation by selecting particular populations displaying greater variation that may contain required medicinal compounds,and by sampling individuals in a population at >20 m spatial intervals to avoid collecting individuals with identical or similar genotypes.  相似文献

We analyzed a bird community in a secondary forest and the results show that the magpie was one of the key groups in the secondary forest. The key group is identified based on the nests used by other birds at a rate of 25%–40.17%. Many species of migratory birds migrate in a series of solitary nocturnal flights. Between flights, they stop to rest and refuel for the next segment of their journey. The mechanism controlling this behaviour has long remained elusive. Here, we show that wild-caught migratory redstarts (Phoenicurus phoenicurus) are consistent in their flight scheduling. An advanced videographic system enabled us to determine the precise timing of flight activity in redstarts caught at a northern European stopover site during their return trip from Africa. Birds were held captive for three days in the absence of photoperiodic cues (constant dim light) and under permanent food availability. Despite the absence of external temporal cues, birds showed clear bimodal activity patterns: intense nocturnal activity alternating with diurnal foraging and resting periods. The onset of their migratory activity coincided with the time of local sunset and was individually consistent on consecutive nights. The data demonstrate that night-migrating birds are driven by autonomous circadian clocks entrained by sunset cues. This timekeeping system is probably the key factor in the overall control of nocturnal songbird migration.  相似文献

Oriental river prawn, Macrobrachium nipponense, is a commercially important freshwater prawn species in China, Japan, Korea and Vietnam. Due to overfishing for food, the wild stocks M. nipponense are endangered. Twenty microsatellite loci were isolated from the M. nipponense. Twelve of these loci were polymorphic (seven to 16 alleles per locus), with expected heterozygosity ranging from 0.68 to 0.86 (n = 48). These polymorphic loci provide a valuable tool for assessing genetic diversity of wild and cultured populations.  相似文献

Chimpanzee populations are diminishing as a consequence of human activities, and as a result this species is now endangered. In the context of conservation programmes, genetic data can add vital information, for instance on the genetic diversity and structure of threatened populations. Single nucleotide polymorphisms (SNP) are biallelic markers that are widely used in human molecular studies and can be implemented in efficient microarray systems. This technology offers the potential of robust, multiplexed SNP genotyping at low reagent cost in other organisms than humans, but it is not commonly used yet in wild population studies. Here, we describe the characterization of new SNPs in Y-chromosomal intronic regions in chimpanzees and also identify SNPs from mitochondrial genes, with the aim of developing a microarray system that permits the simultaneous study of both paternal and maternal lineages. Our system consists of 42 SNPs for the Y chromosome and 45 SNPs for the mitochondrial genome. We demonstrate the applicability of this microarray in a captive population where genotypes accurately reflected its large pedigree. Two wild-living populations were also analysed and the results show that the microarray will be a useful tool alongside microsatellite markers, since it supplies complementary information about population structure and ecology. SNP genotyping using microarray technology, therefore, is a promising approach and may become an essential tool in conservation genetics to help in the management and study of captive and wild-living populations. Moreover, microarrays that combine SNPs from different genomic regions could replace microsatellite typing in the future.  相似文献
